What's the difference between tax exemptions and unchecking collect taxes?

What is the difference between unchecking the collect taxes option or entering a reseller exemption when managing Tax exemptions.

Example: I have a customer from Florida with a valid Reseller's Certificate

Do I uncheck the collect taxes option so no taxes are collected, but there is also an option to enter a Florida reseller's exemption, would this only exempt this customer from sales being delivered to Florida?
