How are carrier shipping rates calcuated.

I am currently starting a website to sell hot sauce in 12oz and 32oz bottles. When calculating the carrier shipping rates does it assume everything will fit in one box? For example, if an order cannot fit in one box how is this calculated during the customer checkout? Is it aware that it must calculate it as multiple packages?
