Accepting credit cards, warehouses, and shipping and fulfilling orders

Some suppliers want only integration with Shopify for US orders / shipping. They don't want international shipping / integration like printed mint app.
Can we create on our shopify account a specific shipping profile for this supplier ie printed mint and set it to us US shipping only, and run our other shipping profiles and suppliers as US and International.

Yes, your approach is perfect. You can create a separate profile for the supplier's products and set up shipping only for the US zone. This way, only customers from the US can purchase the product.
If a customer adds the product to their cart and then changes the destination country at checkout to a non-US location, Shopify will prompt them to remove the item from the cart, as shown in the image below.
