I have an existing website with a homegrown product builder that allows my clients to very specifically customize their product including things like manufacturer logos (from a library within the builder), client uploaded artwork, client uploaded fonts, etc. I'm strongly considering moving my site to Shopify and I'm wondering if there is a way to integrate my existing purpose-built builder? In my head, this is some kind of API integration where the builder passes all relevant information to Shopify where it is added to the cart for purchase.

Seems you want to integrate your existing customization module changes in Shopify.
You have to be specific when you to implement like what kind of changes you will do and where if will reflect.
Shopify provides lots of rest and graphql API to integrate.

As you can see, it includes all kinds of item options, customization options, artwork and design options, etc. In the end, what is created is essentially a custom product with custom options at a specified price. I would like to keep this functionality intact and then pass that custom product and price to Shopify for purchase and fulfillment.
Does this make more sense? Sorry I didn't put it this way to begin with.

I checked all the features of your builder, except the feature of resize the text by user as design.
Everything can be done in Shopify, for designing Shopify has an app customily, request you to test the same.
