I would like to have the header video automatically playing

You’ll need to modify the theme’s code.

Simply add the autoplay attribute to the <video> tag, like this

I have placed it at the bottom of the (video.liquid) section in my code, however it doesn’t seem to be working.

Have a placed the code in the right place?

  1. Upload the video to the Shopify File

  1. Open Theme Setting > Add section > Custom Liquid

  1. Add the below custom code to the Custom Liquid field (let’s add your video URL to replace my demo URL)
