I just started my store and have over $20k in sales. I have register with my state to collect sales tax, but when I go to configure Shopify Tax - I can’t activate it. The button is greyed out. What am I missing?

I’d suggest contacting support. Getting a response from Shopify in here is hit and miss. When you say that you go to configure tax, are you in settings where you can turn it on by state?
