25% discount on full price and 50% on sale items

I need to run a promotion this week.  Instead of using a discount code, I want the discount to be automatically applied at checkout.  I want an additional 50% off discount to be applied on already markdown products at checkout and all other full price products to receive 25% off at checkout. 
Can it be possible?
